Cependant, dans les cas o\u00f9 les valeurs n&#8217;ont pas de sens \u00e9vident, ou si elles varient consid\u00e9rablement, le d\u00e9pannage peut \u00eatre complexe. Il est donc utile d&#8217;\u00e9liminer d\u00e8s le d\u00e9part les sources d&#8217;erreur possibles et de pr\u00eater une attention particuli\u00e8re aux consid\u00e9rations suivantes lors de la configuration et de la r\u00e9alisation des mesures photom\u00e9triques. <\/p>\n\n<p class=\"wp-block-paragraph\"><strong>Installation et environnement<\/strong><\/p>\n\n<p class=\"wp-block-paragraph\"><strong>Solvant <\/strong><br\/>L&#8217;absorbance d&#8217;un \u00e9chantillon est influenc\u00e9e par son pH et la force ionique de son solvant [1]. Des tampons \u00e0 faible teneur en sel et \u00e0 pH neutre ou l\u00e9g\u00e8rement alcalin, qui \u00e9vitent les variations de pH, sont recommand\u00e9s. Des dilutions de l&#8217;ordre de 1:10 sont id\u00e9ales. Les dilutions sup\u00e9rieures \u00e0 1:50 ne sont pas recommand\u00e9es en raison du risque croissant d&#8217;impr\u00e9cisions introduites par cette \u00e9tape de pipetage. Il est donc important de bien m\u00e9langer la solution d&#8217;\u00e9chantillon avant et apr\u00e8s chaque \u00e9tape de dilution, ainsi qu&#8217;imm\u00e9diatement avant la mesure.
